Abstract:Ultraviolet B (UVB) was reported to cause oxidative stress, DNA damage and royal jelly (RJ) was informed to exhibit antioxidant feature in some previous studies. In this study, for the first in the literature, treatment effectiveness of different doses of RJ was investigated against experimentally formed UVB damage. For this purpose, changes which occurred in oxidative stress, genotoxicity and apoptosis were evaluated together with stereological examination. Five equal groups were constituted each including 6 Wistar albino rats. These groups were named as control (C), UVB, RJ 150 mg/kg (RJ150), UVB+RJ 100 mg/kg (UVB+RJ100) and UVB+RJ 200 mg/kg (UVB+RJ200). The animals of UVB, UVB+RJ100 and UVB+RJ200 groups were exposed to UVB radiation for 2 hours once a day during 30 days. RJ was given to rats of RJ150, UVB+RJ100 and UVB+RJ200 groups at 150, 100 and 200 mg/kg doses respectively. All animals were sacrificed on the 30th day. Blood and skin samples were taken for analysis. TOS level of group UVB was significantly (p<0.01) higher compared to all other groups. Marked (p<0.01) decreases of TOS were determined in RJ treated groups comparing to UVB group. When compared with C group, TAS level decreased dramatically (p<0.01) in UVB group whereas significantly (p<0.01) increased again in RJ150 group. With regard to OSI value, its significant (p<0.001) increase was seen in UVB group compared to C and its marked (p<0.001) decrease was seen in RJ150 and UVB+RJ200 groups compared to UVB group. When apoptotic image of our study is analyzed, less DNA fracture was seen in C and RJ150 groups whereas increased DNA fracture was seen in groups where UVB damage was formed. Decrement of DNA fracture was determined in UVB+RJ100 and UVB+RJ200 groups compared to UVB. Comet assay analyze result of UVB group was significantly (p<0.001) higher than those of all other groups. This result was markedly (p<0.001) lower in all RJ treated groups compared to UVB group. When stereological values (volume fraction) were evaluated, a dramatical (p<0.001) increment was seen in UVA group compared to C. This study revealed that RJ could be beneficial in the treatment of UVB damage. Further studies are needed to verify results of this study.
