Survey studies were conducted in 2018 in order to detect tobacco viruses (TMV, TEV, PVY, CMV and TVBMV) in tobacco production areas of Adıyaman province. Multiplex RT-PCR method was used to test the presence of tobacco viruses. Four hundred fifty fresh leaf samples were collected from tobacco fields in Adiyaman province. Of the 450 tobacco samples tested, 40 had CMV (8.8 %), 25 had TMV (5.5 %), and 1 had TMV + CMV infection (0.2 %). The overall infection rate of tobacco viruses in Adiyaman province was found to be 14.4%.According to the molecular tests, TEV, PVY, TVMBV viruses were not detected in the collected tobacco samples. Among the viruses detected in Adıyaman province, all of the coat protein genes of two randomly selected isolates (TMV 20, TMV 307, CMV 61and CMV 73) were cloned and molecularly characterized. TMV Adıyaman isolates showed similarity between 72.3 % and 99.17 % while CMV Adıyaman isolates showed similarity between 74.09-99.54 % at nucleotide level with other 19 different isolates in the World. Othervise, CMV Adıyaman isolates showed similarity between 74.09 % and 99.54 % at nucleotide level with other 20 different isolates in the world. In this study, CMV and TMV were first reported in tobacco production areas of Adiyaman province.
