Abstract:

In some earlier studies, while ultraviolet A (UVA) radiation was reported to cause oxidative stres and DNA damage, caffeic acid phenethyl ester (CAPE) and erdosteine (ERD) were informed to exhibit antioxidant feature. In this study, for the first in the literature, treatment effectiveness of CAPE and ERD was investigated against experimentally formed UVB damage. For this purpose, changes which occurred in oxidative stress, genotoxicity and apoptosis were evaluated together with stereological examination. Four equal groups were constituted each including 6 Wistar albino rats. These groups were named as control (C), UVA, UVA+ERD and UVA+CAPE. The animals of UVA, UVA+ERD and UVA+CAPE groups were exposed to UVA radiation for 2 hours once a day during 30 days. ERD was orally given to animals of UVA+ERD group at a dose of 10 mg/kg 30 min before each UVA application. CAPE was intraperitionally (i.p.) given to animals of UVA+CAPE group at a dose of 10 µmol/kg 30 min before each UVA application. All animals were sacrificed on the 30th day. Blood and skin samples were taken for analysis. TOS level of UVA group was significantly (p<0.01) higher compared to that of all other groups. While TAS dramatically (p<0.01) decreased in UVA group compared to C, its level significantly (p<0.01) increased again in UVA+CAPE group. With regard to OSI value, its significant (p<0.001) increase was seen in UVA group compared to C and its marked (p<0.001) decrease was seen in UVA+ERD and UVA+CAPE groups compared to UVA group. When apoptotic image of our study is analyzed, less DNA fracture was seen in C group whereas increased DNA fracture was seen in groups where UVA damage was formed. Decrement of DNA fracture was determined in ERD and CAPE treated groups compared to UVA group. Comet assay analyze result of UVA group was significantly (p<0.001) higher than those of C but this result was markedly (p<0.001) lower in UVA+ERD and UVA+CAPE groups compared to UVA group. When groups were compared with each other in terms of stereological values (volume fraction), increment was seen in UVA group compared to C and decrement was seen in this group compared to RJ treated groups. This study revealed that ERD and CAPE could be beneficial in the treatment of UVA damage. Further studies are needed to verify results of this study.
