ABSTRACT
Objective: Mespilus germanica L. (Rosaceae) is a medicinal plant used in the treatment of various diseases as well as being consumed as a
fruit among the people due to the abundant polyphenols and antioxidants it contains. The p53 protein produced from the TP53 gene in
humans plays an important role in a wide variety of cell signaling mechanisms that lead to cell cycle arrest, cellular senescence, DNA
repair, differentiation and cell death (apoptosis). Although there are publications on the phytochemical contents and antioxidant activities
of Mespilus germanica in the literature, there is no study on gene expressions in cancer cell lines.
Material-Methods: In this study, various extracts prepared with different solvents from the leaves of Mespilus germanica collected in
different vegetation periods were applied to MCF-7 breast cancer cell lines and their effects on p53 protein expression were investigated.
The extracts were prepared at different concentrations (200, 400, 600, 800, 1000 and 1200 µg/ml) to determine cell viability, dissolved in
DMSO (≤0.005) and applied to MCF-7 breast cancer cell lines for 48 hours. After 48 hours, IC50 and proliferative concentrations were
determined and total mRNA and cDNA synthesis were performed. P53 protein expression level of cDNAs was determined by RT-qPCR.
Results: It was determined that TF coded extract, from many extract types studied, increased P53 protein expression 2-fold in the
proliferative group compared to the control group, and increased P53 protein expression 26-fold in the IC50 group.
Conclusion: Our project, in which we investigated the effect of the p53 gene on the internal mechanisms, continues and the Results will be
