Isolation, Identification and Molecular Characterization of Streptomyces Species from Sediment Samples of the Aras River

Özet

Aras river has a very important place in terms of geographical and ecological sense. It

is thought that the detection, isolation, diagnosis and characterization of Streptomyces diversity

in the Aras river will contribute to the scientific world in the fields of microbiological and

molecular biology and genetics. Streptomyces species have a very important place for the

scientific world, because there are many important substances produced by these bacterial

species, especially antibiotics. Samples taken from Erzurum Horasan, Kars Karakurt and Iğdır

Tuzluca regions where Aras river passes were examined in terms of Streptomyces diversity. The

pH and moisture ratios of the sediment samples taken from the determined localities were

determined. Isolation, purification and color group determination studies were performed using

Bennet's Agar and M65 media. Biochemical, phenotypic and molecular tests were applied to

diagnose these bacterial groups. Spore chain morphology of the isolates determined according to

the color group was visualized (SEM). Sequence analysis of 16 S rDNA gene regions of bacteria

to be isolated microbiologically was performed. Then, the obtained sequences were compared

with the GenBank data, and the molecular affinity and similarity ratios of the isolated bacteria

were obtained.
