The effect of acrylamide given in ovo on the development of the tibia in the embryonic period

Özet
Background of the Study: Acrylamide is a water-soluble monomer used in the synthesis of polyacrylamides. Polyacrylamide is widely used in drinking and wastewater treatment, paper production, crude oil processing, mineral, mine and asphalt production, and soil and sand treatment. Acrylamide is also formed spontaneously during high-temperature frying, baking and roasting of carbohydrate-rich foods. Therefore, some food products prepared for human and animal consumption contain high levels of acrylamide. Additionally, acrylamide can cross the placental barrier and pass into the egg in poultry. For this reason, humans, mammal animals and avians can be exposed to this chemical throughout their lives, starting from the beginning of the embryonic period, and their embryonic development can be negatively affected. Experimental studies have shown that acrylamide has carcinogenic, neurotoxic and genotoxic effects. Chicken’s embryos are a frequently preferred as a material in experimental research to evaluate the possible embryotoxic,genotoxic and teratogenic effects of chemicals, drugs, toxins and many food additives.
Aim: This study aimed to determine the effects of acrylamide injected into fertile chicken eggs on the embryonic development of the tibia.
Method: 390 fertile chicken eggs were used in the study. Eggs were divided into 5 groups as control (n=35 eggs), vehicle control (n=50 eggs), 0.5 μg acrylamide/egg (n=75 eggs), 50 μg acrylamide/egg (n=110 eggs) and 500 μg acrylamide/egg (n=120 eggs) groups. Injections were performed by using sterile insulin syringes. By puncturing the egg with aspecial egg driller, a volume of 100 μl of test solution was injected into the egg yolk and the hole was immediately closed with melted paraffin. Injection were performed immediately before incubation. The eggs were incubated under optimal conditions in the incubator at Selçuk University Faculty of Veterinary Medicine. The right tibiae of 6 embryos and 6 chicks from each group were taken on the 10th, 13th, 17th days of incubation, and on the 2st day (hatching day). The collected tibiae were weighed (g) and their length (mm) was measured.
Results: As a result of the measurements, it was determined that the decreases in tibia length and weight in the acrylamide-administered groups were statistically significant compared to the control and vehicle control groups (P<0.05).
Conclusions: It was concluded that acrylamide given in ovo adversely affected the development of the tibia during the embryonic period.
