Sitotoksik konsantrasyonda glukoz uygulanan NRK-52E hücre hattında glutatyonun nekrotik ve otofajik yolaklara etkisi

Özet
Bu çalışma, diyabetik stres oluşturmak amacıyla sitotoksik düzeyde glukoz uygulanan böbrek hücre hattında, diyabete bağlı komplikasyonların gelişiminde önemli rol oynayan prooksidan/antioksidan dengenin glutatyon uygulamasıyla nekrotik ve otofajik yolaklar üzerindeki etkilerini incelemek amacıyla planlanmıştır. Ayrıca elde edilen bulgular doğrultusunda glutatyonun diyabetik komplikasyonların azaltılmasındaki olası koruyucu etkisinin değerlendirilmesi hedeflenmiştir. Çalışmada NRK-52E hücre hattı in vitro koşullarda kültüre edilmiştir. Glukozun IC50 değeri ile glutatyonun proliferatif etkili dozunun belirlenmesi amacıyla MTT canlılık analizi gerçekleştirilmiştir. MTT canlılık testine göre; glukozun sitotoksik dozu 250 Mm, glutatyonun proliferasyonu artıran dozu ise 250 μM olarak tespit edildi. Deney grupları; kontrol, yüksek glukoz, glutatyon ve yüksek glukoz+glutatyon olacak şekilde oluşturulmuştur. Yirmi dört saatlik inkübasyon sürecinin ardından hücreler toplanmış ve Trizol yöntemi kullanılarak RNA izolasyonu yapılmıştır. Elde edilen RNA örneklerinden cDNA sentezlenmiş, ardından nekrotik ve otofajik yolaklarla ilişkili genlerin ekspresyon düzeyleri RT-qPCR yöntemiyle analiz edilmiştir. Analiz sonuçlarında, glutatyon uygulanan grupta otofajik ve nekrotik gen ekspresyonlarında belirgin bir değişiklik gözlenmezken, yüksek glukoz uygulanan gruplarda bu genlerin ekspresyon düzeylerinin arttığı belirlenmiştir. Bununla birlikte glutatyon uygulamasının, artmış gen ekspresyonlarını azaltarak kontrol grubuna yaklaştırdığı tespit edilmiştir.
This study was designed to investigate the effects of glutathione administration on necrotic and autophagic pathways in a renal cell line exposed to cytotoxic concentrations of glucose to induce diabetic stress, with particular emphasis on the prooxidant/antioxidant balance, which plays a critical role in the development of diabetes-related complications. Furthermore, the study aimed to determine whether glutathione has a significant protective effect in reducing diabetic complications based on the obtained findings. NRK-52E cells were cultured under in vitro conditions. The IC50 value of glucose and the proliferative dose of glutathione were determined using the MTT cell viability assay. According to the MTT viability test, the cytotoxic dose of glucose was determined to be 250 μM, and the proliferation-enhancing dose of glutathione was also determined to be 250 μM. Experimental groups were established as control, high glucose, glutathione, and high glucose + glutathione. Following 24 hours of incubation, the cells were collected and RNA isolation was performed using the Trizol method. Subsequently, cDNA synthesis was carried out, and the expression levels of necrotic and autophagic genes were analyzed by RT-qPCR. According to the results of the analysis, no significant alteration was observed in the expression levels of autophagic and necrotic genes in the glutathione-treated group. However, these genes were upregulated in the high-glucose groups. In addition, glutathione treatment reduced the increased gene expression levels and brought them closer to those observed in the control group.
