Özet

The aim of this study was to evaluate the chemical characterization and the anti-ageing and anti-inflammatory activities of extracts prepared from the aerial part of Nepeta betonicifolia CA Meyer. The plant material was harvested from Gevaş district, Van province, Turkey, in June 2021. The aerial parts of the plant were macerated separately with n-hexane, ethyl acetate and methanol solvents. The tyrosinase, elastase and cyclooxygenase (COX) inhibitory activities of the extracts were tested in vitro. Phytochemical analysis of the extracts was conducted using liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) with 56 standards.. All samples significantly inhibited the tyrosinase enzyme at concentrations of 100-500 µg/mL as follows: n-hexane extract, 46.78±0.35-68.57±1.95%, methanol extract, 28.03±3.30-84.54±3.25%, and ethyl acetate extract, 27.34±1.45-97.50±2.50%. While the extracts showed no inhibitory effect on elastase at a concentration of 100 µg/mL, the inhibitory effect of the extracts at a concentration of 500 µg/mL was as follows; methanol extract, 46.33±3.26%, ethyl acetate extract, 59.55±4.87%, and n-hexane extract, 94.95±1.15%. The all extracts showed less than 5% inhibitory effect on COX (cyclooxygenase) enzymes at a concentration of 100 µg/mL. The main compounds detected in the extracts were cosmosiin, rosmarinic acid, luteolin-7-glucoside, chlorogenic acid and quinic acid. To our knowledge, the inhibitory effect of Nepeta betonicifolia on tyrosinase, elastase and COX enzymes has been reported for the first time in this study.
