Lipozomlar ilk defa İngiliz hematolog Dr. Alec D. Bangham ve arkadaşları tarafından 1961
yılında keşfedilmiştir. Yapılarının temel elemanı fosfolipidlerdir. Fosfolipitlerin sahip olduğu
özellikler sayesinde hidrofilik ve hidrofobik uçları sulu sistemlerle karşılaştıklarında hidrofobik
uçların sudan kaçma eğilimi sonucu çift moleküllü tabakalar halinde dizilerek dairesel halkalar
oluşturur. Bu dairesel halkalara lipozom adı verilir. Lipozomların yapısında; tek veya çok
tabakalı bir membran, orta iç kısımda sulu bir boşluk vardır. Buna göre lipozomlar yağda
çözünen maddeleri lipozom zarında, suda çözünen maddeleri de orta iç kısmında taşıyabilir. Bu
sayede lipozom zarına hapsolmuş olan madde, sulu faz ile; sulu faza hapsolmuş madde de
hidrofobik faz ile etkileşime girmemekte ve etken madde özelliğini koruyabilmektedir.
Amfipatik yapıya sahip olan lipozomlar içerdikleri hidrofilik ve hidrofobik bölgeler nedeniyle
etken maddeleri hedef bölgeye kontrollü olarak taşıyabilme, serbestleştirebilme ve biyolojik
olarak parçalayabilme özelliğine sahiptir. Lipozomların yapıları hücre membran yapısına
benzediği için ilk olarak hücre membranı ile ilgili çalışmalara konu olmuştur. Hücre zarına
benzerlik göstermeleri, toksik olmamaları, kimyasal yapılarının araştırmacılar tarafından
belirlenebiliyor olmasından dolayı bilim adamları tarafından uzun yıllardır model membran
olarak kullanılmıştır. Deneysel koşullarda sentetik fosfolipitlerin değişik yöntemlerle sulu bir
ortama yayılması sonucu oluşan lipozomlar, hazırlama aşamasında dışarıdan eklenen diğer
maddeleri de enkapsüle edebilme özelliğine sahip oldukları için kullanımları giderek
artmaktadır. Lipozomlar eczacılık, kozmetik, gıda, deterjan, tekstil ve bunun gibi birçok alanda
uygulama potansiyeline sahiptir. Lipozomların gıdalarda kullanımı halen gelişmekte olan bir
alan olup gıda bilimciler lipozomları proteinler, enzimler, vitaminler ve aromalar gibi
fonksiyonel bileşenlerin kontrollü salınımı için çeşitli gıda uygulamalarında kullanmaya devam etmektedirler. Bu çalışmada lipozomların genel özellikleri, hazırlama yöntemleri,
sınıflandırılmaları ve gıda alanında kullanımları derlenmiştir.
Liposomes were first discovered by British hematologist Dr Alec Bangham and his friends in
1961. The basic elements of their structure are phospholipids. Thanks to the properties of
phospholipids, hydrophilic and hydrophobic ends when they encounter aqueous systems, the
hydrophobic ends tend to escape from water, forming double-molecular layers and forming
circular rings. These circular rings are called liposomes. In the structure of liposomes; a single
or multi-layer membrane, in the middle there is a watery void. Accordingly, liposomes can
carry fat-soluble substances in the liposome membrane and water-soluble substances in the
middle interior. Thus, the substance trapped in the liposome membrane, with the aqueous phase;
The substance trapped in the aqueous phase also does not interact with the hydrophobic phase
and can preserve its active ingredient properties. Liposomes, which have an amphipathic
structure, have the ability to carry active substances to the target area in a controlled manner,
release and biodegrade due to the hydrophilic and hydrophobic regions they contain. Since the
structure of liposomes resembles the cell membrane structure, it was first subject to studies on
the cell membrane. It has been used as a model membrane by scientists for many years because
of its resemblance to the cell membrane, non-toxicity, and the fact that its chemical structures
can be determined by researchers. Liposomes, which are formed as a result of spreading
synthetic phospholipids into an aqueous environment by different methods under experimental
conditions, are increasingly used because they have the ability to encapsulate other substances
added externally during the preparation phase. Liposomes have application potential in many
areas such as pharmacy, cosmetics, food, detergent, textile and so on. The use of liposomes in
food is still an emerging field, and nutritional scientists continue to use liposomes in a variety
of food applications for the controlled release of functional ingredients such as proteins,
enzymes, vitamins and flavors. In this study, general properties, preparation methods,
classification and food use of liposomes are compiled.
